چکیده

In all of ten strains of H. influenzae examined prior to exposure to streptomycin a very small fraction of the bacteria formed colonies in the presence of 1,000 units of streptomycin per cc. These variant organisms possess the characteristics of bacterial mutants. 1. Different independent cultures of the same strain on different occasions, and even cultures seeded in a single test with the same inoculum, show marked variation in prevalence of resistant variants in populations of comparable size. This variation is just as great in genetically homogeneous culture sources as in heterogeneous sources. Evidence is presented for the continuous random occurrence of these resistant organisms. 2. The rate of occurrence of the resistant members is not significantly different for the ten strains studied; it varies from 2.6 x 10(-11) to 7.0 x 10(-11) per bacterium per bacterial generation. The results of five different tests on the same strain show comparable variation, 2.9 x 10(-11) to 5.3 x 10(-11). This low rate contributes evidence consistent with the mutation hypothesis. 3. Variants exhibiting resistance to 1,000 units of streptomycin per cc. transmit this trait unchanged in degree through many generations. The results obtained after twelve or more subcultures in streptomycin-free media suggest that in a portion of the colonies resistant to streptomycin an additional trait, differing from those exhibited by the parent culture, is associated; the nutritional requirements of these cultures are different.

Induction of Streptomycin Resistance in Sensitive Hemophilus Influenzae by Extracts Containing Desoxyribonucleic Acid from Resistant Hemophilus Influenzae

Resistance to streptomycin, of a degree exceeding 1000 microg. per ml., has been induced in sensitive strains of Hemophilus influenzae by exposure for 10 minutes to desoxyribonucleic acid-containing extracts isolated from a strain of type b Hemophilus influenzae which had emerged resistant to 1000 microg. of streptomycin per ml. DNA is essential for the process which brings out this change; the...

Studies on the Nature of Hemophilus Influenzae Cells Susceptible to Heritable Changes by Desoxyribonucleic Acids* by Hattie E. Alexander, M.d., Grace Leidy, and Eros Hahn

There is now convincing evidence that highly specific desoxyribonucleic acids (DNA's) can direct some inherited traits of members of 3 different bacterial species, Diplicoccus pneumoniae (1), Hemophilus influenzae (2, 3), and Neisseria rnenlngitidis (4), and suggestive evidence that heritable traits of certain viruses, rabbit fibroma (5), and bacterial viruses (6), may be influenced by the acti...

In Vitro Production of New Types of Hemophilus Influenzae

Two new types of Hemophilus influenzae, Sab and Sad have been produced in vitro. Each exhibits the presence of the type specific polysaccharides of 2 types of E. influenzae within the same cell. In Sab the polysaccharides of types a and b have been demonstrated and in Sad those which characterize types a and d. The Sab and Sad traits are inherited. Sab was produced by the action of DNA-containi...

Single variant bottleneck in the early dynamics of H. influenzae bacteremia in neonatal rats questions the theory of independent action.

There is an abundance of information about the genetic basis, physiological and molecular mechanisms of bacterial pathogenesis. In contrast, relatively little is known about population dynamic processes, by which bacteria colonize hosts and invade tissues and cells and thereby cause disease. In an article published in 1978, Moxon and Murphy presented evidence that, when inoculated intranasally ...

Methylase activities from Haemophilus influenzae that protect Haemophilus parainfluenzae transforming deoxyribonucleic acid from inactivation by Haemophilus influenzae endonuclease R.

Specific methylases that have the properties of deoxyribonucleic acid (DNA) modification enzymes have been isolated from Haemophilus influenzae strain Rd. Two activities ((Methylase IIa and methylase III) were found to protect transforming DNA of H. parainfluenzae from the action of H. influenzae restriction enzymes. To determine the specificty of the protection, a procedure based on biological...
